The Nikon LiveScan Swept Field Confocal Microscope (SFC) enables dynamic live cell confocal imaging. Its revolutionary new optical design is used in point scan mode for high resolution data acquisition at frame rates exceeding 200 frames/sec, and in slit scan mode at acquisition frequencies exceeding 1500 frames/sec, while minimizing specimen exposure to damaging illumination energy. For cell biologists and neurophysiologists, Nikon's LiveScan SFC microscope can monitor and record rapidly occurring events in msec time resolution within living cells without compromising spatial resolution, while simultaneously controlling high frequency, low intensity illumination which substantially reduces photobleaching and phototoxicity. Both the DM-IRBE and the DMLFS are mounted on air tables to prevent image noise caused by vibration. Image stacks generated by the confocal microscope may be archived using a proprietary Leica format or may be exported as multiple TIFF files. All three microscopes can be used as confocal microscopes by changing the Scan Electronics from one scope to the next, a procedure that takes approximately 5 minutes. The confocal microscopes use the Leica TCS/SP Confocal Scanner which is controlled by proprietary Leica software. |

The Confocal Laser Scanning Microscopy (CLSM) Facility is operated in cooperation with the CGRB and the EHSC. A NIH Shared Instrumentation Grant supplemented by the OSU Research Office provided funding for the Zeiss LSM 510 Meta. The facility is open to users around campus. We can provide assisted imaging sessions, or for longer term projects, training. The system is equipped with 3 lasers (Argon and Helium/Neon), which give excitation lines of 488,543, and 633nm. The Leica Confocal Microscope detects photons emitted by the fluorophores using patented technology that allows for limitless emission filter settings. The scan head contains a spectrophotometer which is used to adjust the emission wavelength width, and an acousto-optical tunable filter (AOTF), which is used to control the percentage and mixture of laser light. The galvanometric Z-stage has a maximum travel of 170 microns, and can move in 41 (+/- 10nm) steps. The scan head is controlled by a Pentium II 300MHz computer with Windows NT operating system. |
